IMAGE TOOLBOX 2.4 software allows the user to browse trough the images acquired during a Time Lapse Experiment. It is possible to sample the images and export thumbnail pictures and avi movies.
Image Toolbox 2.4 fully interact with Oko-Vision 2.7. In case a Time Lapse Experiment is running, to check in real time the progress of the experiment, the operator can synchronise the data between the two software from time to time by clicking on the Update button.

The operator can choose to make an avi movie where images are mounted one after the other, it is possible to make an avi movie of a selected layer of a specific object along the time, or it is possible to make an avi movie where the different layers of a specific set of a z stack are mounted one after the other. Before making the avi the operator can choose to subtract the background, if the background was ever acquired during the Time Lapse Experiment. In this case a dedicated algorithm will perform the background subtraction on each image before mounting the avi movie. | ||
MOSAIC This tool allows the operator to build thumbnails. It is possible to display side by side several images. The operator can build three different type of mosaic: Object, Time, Layer by selecting the mode and clicking on the Go button. Time and temperature are reported in each image. 1. Object: The operator can indicate a single set (i.e., fix the time) and indicate a specific Layer (in the case of Z stack) than it is possible to select the objects to visualize. The Mosaic will build a thumbnail image placing side by side images of all the objects at the specific set and layer. 2. Time: The operator can indicate a single object and indicate a specific Layer (in the case of Z stack) than it is possible to select the set (i.e., the time) to visualize. The Mosaic will build a thumbnail image placing side by side images at different times of the specific object and layer. 3. Layer: The operator can indicate a single object and indicate a specific set (i.e., fix the time) than it is possible to select the layers to visualize. The Mosaic will build a thumbnail image placing side by side images at different layers of the specific object at the selected time. Once made the mosaic, the operator can Save the thumbnail image by ckicking on the Save button. It is also possible to extract the images: in this case all the images, placed in the thumbnail, will be separately copied in the destination directory and saved as image0001.tif, image0002.tif, etc. |

CHANNELS This tool allows to merge or split different fluorophore signals. Furthermore, for any single channel, operations described above can be performed. |

MEASUREMENTS This module allows the user to perform geometric and intensity measurements on the image, image histogram, linear intensity profile, on each fluorescent channel. Automatic reiteration of the measurement on the entire experiment allows easy analysis of dynamic phenomena and quantitative comparison of images. |

CONTRAST EQUALIZER This module allows the user to perform LUT enhancement on each individual channel. Automated routines are available to perform background subtraction and best LUT equalization. The Auto LUT equalization function implements an algorithm that will compensate for light intensity fluctuation or progressive fluorescence bleaching. |
